Promo is when you give records away free to promote your track
Label release is when the label you are signed up to releases your record to the public

Labels always promo out their new tracks to major DJs for reactions and charting (search Cool Cuts), this not only helps them to get their track listened to by the masses due to the DJs residencies/radio slots/gigs, but also gives the label a good idea of how well their track will do.
